Mismagius has the Speed and Special Attack to use Calm Mind effectively, and three immunities make it quite easy to bring in to play. The idea is simple: Calm Mind up, from behind a Substitute, and attempt a sweep. Alternatively, you can burn any physical attackers that switch in, making Mismagius difficult to take down from either side of the attacking spectrum. Taunt is a big help against phazers and other stat uppers, like Calm Mind Blissey and Suicune.

With Substitute, Hidden Power Fighting, and Leftovers, you can beat Blissey 1v1 and proceed to sweep their team. Colbur Berry helps against Weavile that might switch in during a Calm Mind or a Shadow Ball, allowing you to either kill or Burn them. While Hidden Power Fighting is optimal on this set, Thunderbolt can work if you're unable to obtain it. If you use Substitute, use it with Leftovers.

Mismagius is one of the few Perish Trappers, and is the best defensively. Mean Look something, use Perish Song, then Protect for the next two turns to prevent damage. Taunt is to stop Whirlwind / Roar from ruining your strategy, and Substitute if you're faster and don't want to rely on Protect's 50% chance of working the second time. Pain Split can work as a makeshift Recover, since you aren't doing any damage to them, and hopefully aren't wasting your time Perish Songing an enemy with low HP.

You can use a special attack in the last slot too. Taunt Gyarados will shut you down, and Skarmory can phaze away Mean Look, so you can pack Thunderbolt to deal with them. Hidden Power Fighting can bring down Weavile and Tyranitar, who can both kill Mismagius before it gets the chance to stall them out with Perish Song.

Gengar is generally a better option for this type of set, due to a superior movepool and higher Speed and Special Attack. Still, Mismagius has better defensive abilities, which makes for easier switch-ins. Energy Ball is mostly filler, although it does make it easier to take down Ground types. Destiny Bond and her great Speed means Mismagius can take out a Swords Dance Garchomp, or another slower foe that she is unable to KO in a single hit.

Burn physical attackers, Paralyze special sweepers and plug away with your attacking moves. A really simple set, but very effective nonetheless. Mismagius' three immunities are a big help here, as it helps to get her in so she can spread status around.

Confuse Ray can be used over either status move or your secondary attack for further annoyance. It works particularly well with Thunder Wave. Pain Split is also an option as it will annoy Blissey and Snorlax no end.

Other Options

Spite might help a team mate set up if you can drain all the PP of an opponents main attacking move, but the above sets cover Mismagius's viable options.

EVs

Take advantage of the 105 base Speed. A Pokémon with Will-O-Wisp or Destiny Bond that can outspeed threats like Garchomp is always handy to have around, so a minimum of 334 Speed is recommended. Besides that, concentrate on Special Attack if you're going with an attacking set, or HP and Special Defense for the more defensive/supportive sets. Will-O-Wisp should cover your physical Defense for the most part.

Opinion

A pure special attacking Ghost seems like a good idea, but Mismagius is hampered by its small, predictable movepool. Still, the dual threat of Calm Mind and Perish Song can catch opponents off-guard, since they have radically different counters. This also means she can cause serious problems for teams who are overly reliant on Blissey to wall special attackers.

Counters

For Special attacking variants: Weavile or Tyranitar if you can get it in on a Calm Mind or Shadow Ball, Houndoom can come in on both of those and Will-O-Wisp. It has 60 base Defense, so Physical attackers with decent Special Defense are the obvious route, but watch out for Will-O-Wisp. There's not much you can do about that but use a Fire type or Guts Pokémon, though, so Mismagius may be doing its job after all.

For Perish Song: Hit it very hard with physical attacks, since you only get around two shots at it. Weavile will instantly kill Perish Songers, since it can switch in without fear and kill it whether it switches or not. Tyranitar can do the same. Choice Band / Choice Specs Pokémon can get the quick damage you need to take it out before it can start Protecting. At least it doesn't get Hypnosis anymore...
