Vileplume's great defensive typing, solid bulk and useful movepool combine to make it a decent defensive option in the tier. Its bulk notably allows it to check many threatening Pokemon that are otherwise difficult to handle, like Cobalion, Rhyperior, Escavalier, and Dragon Dance Haxorus thanks to Strength Sap lowering their Attack, while its typing grants it the ability to switch into and threaten many defensive Pokemon in the tier, like Milotic, Gastrodon, and Sylveon. However, it faces stiff competition from Roserade, which is faster and stronger and has access to Spikes to wear down opposing teams over time. Nevertheless, Vileplume carves a niche through its superior bulk and ability to deal with many of Roserade's checks, like Escavalier and Copperajah, as well as dissuading their physical attacks thanks to Effect Spore. Strength Sap allows Vileplume to both check and reliably recover against many physical attackers, like Pangoro and Doublade. Leech Seed allows Vileplume to break through Pokemon that would otherwise wall its STAB combination, like Escavalier, Doublade, and Cobalion. Sleep Powder can be used instead, allowing Vileplume to incapacitate a check like Incineroar or Noivern, but it leaves it walled by most of the Steel-types in the tier. 84 Speed EVs are used to outspeed cleric Sylveon, allowing Vileplume to check it more reliably, with the remaining EVs going into its physical bulk.

Vileplume fits best on defensive teams that appreciate its ability to handle many of the tier's physical attackers. Despite checking a unique and wide range of Pokemon, Vileplume is still weak to a host of top-tier threats and requires support to handle them. Pokemon like Incineroar, Noivern, and Toxtricity can switch into Vileplume's attacks and threaten it with their STAB moves or, in Toxtricity's case, Boomburst. Rhyperior makes for an excellent partner, checking all three of the aforementioned Pokemon for Vileplume. In return, it appreciates Vileplume's ability to deal with opposing Water- and Grass-types like Milotic and Tsareena. Sylveon is a helpful teammate that can provide Vileplume with Wish support, which is useful, as Vileplume's main source of recovery, Strength Sap, is not reliable on its own. Sylveon can also help with checking Pokemon like Noivern and Choice Band Haxorus that Vileplume struggles with. In return, Sylveon appreciates Vileplume's ability to deal with Pokemon like Cobalion, Escavalier, and Copperajah, and can run Heal Bell more freely as a result.
